摘 要: 肺纤维化是一种慢性、进行性、纤维化性间质性肺炎,发病机制不明,预后极差。目前已建立多种肺纤维化动物模型,对研究肺纤维化的发病过程、形态功能改变及治疗具有重要意义,其中以博来霉素最为常用,并可以选择多样化的给药方式。但大多数研究并未系统阐明各种肺纤维化模型动物间差异,不同模型各有优、缺点。本文主要从药物、环境、细菌内毒素脂多糖、转基因腺病毒载体以及复合因素等多个方面阐述各类模型的构建方法及优、缺点,为后续选择用于肺纤维化药物开发及临床研究的动物模型提供参考。 |

Abstract: Pulmonary fibrosis is a chronic,progressive,fibrotic interstitial pneumonia with unknown pathogenesis and extremely poor prognosis.A variety of animal models of pulmonary fibrosis have been established,which are of great significance for the study of the pathogenesis,morphofunctional changes and treatment of pulmonary fibrosis.Among these models,bleomycin-induced pulmonary fibrosis is the most commonly used,as it allows for a variety of drug delivery options.However,most studies have not systematically elucidated the differences between animals in various pulmonary fibrosis models,and different models have their own advantages and disadvantages.In this paper, we describe the construction methods,advantages and disadvantages of various models from the aspects of drug,environment, bacterial endotoxin,transgenic adenovirus,and composite factors,which can be used as a reference for the subsequent selection of models for drug development and clinical research of pulmonary fibrosis. |
